ABSTRACT

Ribosomes are abundant, large RNA-protein complexes that are the sites of all protein synthesis in cells.Defects in ribosomal proteins(RPs),including proteoforms arising from genetic variations, alternative splicing of RNA transcripts,post-translational modifications and alterations of protein expression level, have been linked to a diverse range of diseases, including cancer and aging.Comprehensive character-ization of ribosomal proteoforms is challenging but important for the discovery of potential disease biomarkers or protein targets. In the present work, using E.coli 70S RPs as an example,we first developed a top-down proteomics approach on a Waters Synapt G2 Si mass spectrometry(MS)system, and then applied it to the HeLa 80S ribosome. The results were complemented by a bottom-up approach. In total,50 out of 55 RPs were identified using the top-down approach.Among these, more than 30 RPs were found to have their N-terminal methionine removed.Additional modifications such as methylation, acetylation, and hydroxylation were also observed, and the modification sites were identified by bottom-up MS.In a HeLa 80S ribosomal sample,we identified 98 ribosomal proteoforms, among which multiple truncated 80S ribosomal proteoforms were observed, the type of information which is often overlooked by bottom-up experiments.Although their relevance to diseases is not yet known, the integration of top-down and bottom-up proteomics approaches paves the way for the discovery of proteoform-specific disease biomarkers or targets.

ABSTRACT

Liver is the central hub regulating energy metabolism during feeding-fasting transition. Evidence suggests that fasting and refeeding induce dynamic changes in liver size, but the underlying mechanisms remain unclear. Yes-associated protein (YAP) is a key regulator of organ size. This study aims to explore the role of YAP in fasting- and refeeding-induced changes in liver size. Here, fasting significantly reduced liver size, which was recovered to the normal level after refeeding. Moreover, hepatocyte size was decreased and hepatocyte proliferation was inhibited after fasting. Conversely, refeeding promoted hepatocyte enlargement and proliferation compared to fasted state. Mechanistically, fasting or refeeding regulated the expression of YAP and its downstream targets, as well as the proliferation-related protein cyclin D1 (CCND1). Furthermore, fasting significantly reduced the liver size in AAV-control mice, which was mitigated in AAV Yap (5SA) mice. Yap overexpression also prevented the effect of fasting on hepatocyte size and proliferation. Besides, the recovery of liver size after refeeding was delayed in AAV Yap shRNA mice. Yap knockdown attenuated refeeding-induced hepatocyte enlargement and proliferation. In summary, this study demonstrated that YAP plays an important role in dynamic changes of liver size during fasting-refeeding transition, which provides new evidence for YAP in regulating liver size under energy stress.

ABSTRACT

@#Adeno-associated virus (AAV) is a common viral vector used in gene therapy.Because of its high safety and its ability to target a variety of cells, it has been widely used in preclinical and clinical studies. However, during the design and production, AAV vectors have many key quality attributes that affect their safety and efficacy. The development and application of biological mass spectrometry technology provides a convenient platform for the research on biological macromolecules, especially in the aspects of protein sequence, structure and interaction. For AAV vectors, mass spectrometry can facilitate the determination or characterization of capsid protein ratio, post-translational modification, serotype, and empty capsid ratio, thus assisting in the quality control of AAV vectors.Compared with the existing methods, mass spectrometry has the advantages of smaller amount of sample size, faster and more sensitive analysis, being more suitable for the analysis of complete AAV vectors with higher mass resolution, and can distinguish empty capsids, full capsids and partial capsids. In the future, mass spectrometry technology is expected to play a more important role in the design and production of AAV vectors through the coupling of more efficient protein separation technology with mass spectrometry, the development of new information processing software platforms and new mass spectrometry detection techniques.

ABSTRACT

Objective To investigate the intervention effect of 1,25(OH)2D3 on serum level of parathyroid hormone, bone metabolic markers and muscle strength in the community elderly with low bone mass. Methods A total of 132 residents aged 60 years and over from Shanghai communities diagnosed as osteopenia or osteoporosis were enrolled and treated with 0.5 μg/d calcitriol for 3 months.Serum levels of parameters in blood biochemistry and bone metabolic markers were determined by Cobas Diagnosis System of Roche before and after treatment, and the handgrip strength of both left and right hands were measured at the same time. Results The average age of 132 patients was(66.1 ± 6.3) years,4 cases (all female) terminated their medication in advance, and 128 patients completed the treatment for three months. The baseline serum levels of 25 (OH)D and PTH were 19.6 ± 7.9 μg/L and 41.0 μg/L respectively.According to the median value of serum PTH before treatment, patients were divided into the two groups:the high PTH group and the low PTH group.Compared with the low PTH group, the serum levels of 25OHD and serum calcium were decreased in the high PTH group[(17.1 ± 7.0) μg/L vs.(22.1 ± 8.2) μg/L,2.4 mmol/L vs.2.5 mmol/L,P<0.05].After 3 months of intervention, serum levels of creatinine, urine calcium and left handgrip were significantly elevated[(68.2 ± 13.8) μmol/L vs.(65.2 ± 13.4) μmol/L,(5.9 ± 2.8)mmol/24 h vs.(4.4 ± 2.0) mmol/24 h,23.8 kg vs.21.0 kg,all P <0.05],while serum levels of PTH,eGFR,phosphorus were significantly decreased[35.5 ng/L vs.42.0 ng/L,(87.0 ± 17.0) ml/min vs.(93.1±17.9) ml/min,1.2 mmol/L vs.1.3 mmol/L,all P<0.05).There was a significant negative correlation between serum 25(OH)D and PTH before treatment(r=-0.312,P<0.05),but the negative correlation between them was no longer significant after 3 months of treatment (r =0.042,P > 0.05).A multivariate logistic regression analysis revealed that the increment of left handgrip strength greater than 25% (OR =0.138,95% CI:-0.002-8.383),the increment of serum calcium levels(OR =2.578,95%CI:1.0345-8.693)and age(OR =0.103,95%CI:0.035-0.345) were significantly correlated with the decrement of serum PTH levels greater than 30 % after three months of treatment. Conclusions Vitamin D diminution or deficiency is common in the elderly. The shortterm treatment of calcitriol can obviously reduce serum PTH,inhibit bone absorption and increase muscle strength. The effect of calcitriol on serum PTH is closely related to promoting calcium absorption and improving handgrip strength.

ABSTRACT

Objective To investigate the prevalence and risk factors of vertebral fractures in the elderlys in Shanghai community. Methods Stratified sampling was performed according to the age distribution of the population in 3 urban and 2 suburban communities in Shanghai, and a total of 2 929 old people aged over 65 years were enrolled. The thoracic and lumbar radiographs and questionnaires were obtained in all the participants. The diagnosis of vertebral fractures were evaluated using Genant′s semi-quantitative method, and the prevalence of vertebral fracture was calculated. Logistic regression was used to analyze the risk factors of vertebral fracture. Results ( 1) The prevalence of vertebral fractures in community-dwelling elderly was 14.4%totally,and 20.1%in individuals above 80 years old, which was significantly higher than the others (P<0.05). The prevalence of vertebral fractures in females was significantly higher than that in males (18.5%vs 12.4%, P<0.05). (2) The most common vertebral fracture sites were T12 and L1. Grade 1 vertebral fracture was most common in our cohort. The proportions of grade 2 and grade 3 were significant higher in females than that in males (30.6% vs 17.0%, P<0.05). (3) Gender and sex had an interaction effect on the prevalence of vertebral fractures. Prevalence of vertebral fractures increased steadily with age in both genders, although the gradient was steeper for women (P<0.001). (4) Logistic regression analysis showed that: female, age, parental hip fracture history, previous fracture history, and alcohol consumption were risk factors of vertebral fractures in community elderly. Smoking, long-term use of glucocorticoid, various diseases associated with secondary osteoporosis had no significant correlation with vertebral fracture. Conclusion The vertebral fractures were more common and serious in women than in men in community elderly. Age was significantly correlated with the presence of vertebral fracture in women than that in men. Female, age, parental hip fracture history, previous fracture history, and alcohol consumption were risk factors of vertebral fractures in community elderlys.

ABSTRACT

To summarize the clinical experience of Professor WANG Meng-yong in the treatment of uric acid nephropathy. Professor WANG believes that the disease is mainly caused by spleen and kidney deficiency, disorder of function of Sanjiao, and pathological products, such as phlegm dampness and blood stasis and other metabolic disorders. Therefore, the treatment should distinguish symptoms and essence. Starting from pathogenesis and pathological features of spleen and kidney deficiency and phlegm dampness and blood stasis, the treatment should flexibly apply the methods of nourishing spleen and kidney, reducing phlegm and dispelling humidity, and activating blood and using diuretic of hydragogue to alleviate water retention, which can greatly reduce side effects caused by the long-term use of Western medicine and the onset of gout, and then to help disease recovery.

ABSTRACT

correlation with cross-sectional area and negative correlation with bucking ratio. The effect of fat mass on hip geometric parameters at the three regions was inconsistent. (3) Fat mass and percentage of body fat ( Fat%) showed negative correlation with cross-sectional area and average cortical thickness and positive correlation with bucking ratio in young group. However, the negative contribution produced by fat mass and Fat% to hip geometric structure became weaker gradually in middle-aged and older groups. ( 4 ) Limb lean mass yielded the largest positive contribution to cross-sectional area and average cortical thickness at all three regions in young group. However, the contribution produced by trunk lean mass became the largest positive contribution to cross-sectional area and average cortical thickness in older group. Conclusions Among all the body composition parameters, hip BMD has showed the most marked correlation with hip geometric structure. Lean mass is ranked the second in the correlation. The impacts of fat mass and lean mass on hip geometry are changing with aging and their different tissue distributions.

ABSTRACT

Objective Rapamycin (RAPA) is an anti-proliferative immunosuppressant and has been used to suppress rejection of transplanted organs. In present study, we observed the effect of rapamycin on epithelial-myofibroblast transition (EMT)of cultured HKC cells in vitro. Methods Cultured human proximal tubular epithelial cells (HKCs) were divided into three groups: blank control, treated with TGF-?1 (1 ?g/L) and treated with TGF-?1 (1 ?g/L) plus rapamycin (0.1, 1, 10, 100 ?g/L). The protein and mRNA for ?-SMA and E-cadherin in HKC cells were determined by Western Blot and RT-PCR.The mRNA level of Snail in HKC was detected by RT-PCR. Results Rapamycin dramatically abrogated TGF-?1 induced ?-SMA expression and restored E-cadherin expressionin HKC cells in a dose-dependent manner. At a concentration of 100 ?g/L, rapamycin almost completely blocked ?-SMA mRNA and protein expression induced by TGF-?1(1 ?g/L). Rapamycin also suppressed expression of ?-SMA in HKC cells at both mRNA and protein level in a time dependent manner. We also found rapamycin dramatically abrogated TGF-?1 induced Snail mRNA expression in HKC cells in a dose-dependent manner. Conclusion Rapamycin may inhibit EMT of tubular cells in vitro. The downregulation of Snail expression might be one of the mechanisms of rapamycin blocking EMT.

ABSTRACT

Objective:To study the relationship between the levels of serum insulin,C-reactive protein(CRP),Tumor Necrosis Factor(TNF-?),insulin resistance index, blood fat,and phlegm-stagnation stasis syndrome of metabolic syndrome(MS),so as to discuss the signifi cance of phlegm-stagnation stasis syndrome in treatment based on syndrome differentiation of MS,in order to fi nd the objective evidence of differentiation of symptoms and signs of TCM of MS.Methods: In this study,54 cases with MS were divided into groups of phlegm-stagnation stasis syndrome and non-phlegm-stagnation stasis syndrome according to the clinical symptoms, tongue demonstration and pulse tracings. There were 25 cases in phlegm-stagnation stasis group,29 cases in non-phlegm-stagnation stasis group. The levels of serum insulin,CRP,TNF-?,blood fat were checked, the insulin resistance index was calculated. Results:The levels of serum insulin,CRP,TNF-? of the two groups of metabolic syndrome were signifi cantly higher than those of the healthy persons(P

ABSTRACT

Objective To evaluate the relationship between osteoporosis(OP) and the age of menarche and menopause, parity and lactation and bone mineral density(BMD).Methods BMD was measured in 1472 postmenopausal women by Norland XR-36 dual energy Xray absorptiometry(DXA) from May 1999 to April 2003 in our hospital. All data of BMD were compared and statistically analyzed between women with different age of menarche and menopause, parity and lactation period. The diagnostic criteria of osteoporosis was defined by a loss of BMD greater than 2.5 standard deviations below average peak BMD of young adult women. Results In all 1472 subjects, the incidence of osteoporsis is 58.5%(861/1472).There was significant increase in osteoporosis of lumbar spine in women with the age of menarche≥17(119 out of 336 cases were with osteoporosis in lumbar)compared with women with age of menarche≤13(75 out of 276 cases were with osteoporosis in lumbar). In women with menarche age of 11-13,14-16,17-19,lumbar vertebral(L_(2-4)) BMD values were(0.83?0.16),(0.82?0.16),(0.80?0.14) g/cm~2,respectively.There was significant increase in BMD of lumbar in the menarche age group of 11-13 compared with the age group of 17-19 in women within menopause 1-10 years(P

ABSTRACT

[Objective] To investigate the effect of Kun'an Granule (KG) on serum hormone level in rats with hyperprolactinemia [ Methods ] Rats were randomized to five groups: group A ( mimic operation) , group B (model control), group C (bromocriptine), group D (low-dose KG) and group E (high-dose KG). Except group A, the other four groups were implanted with pituitary for modeling. Group C was treated with bromocriptine 1 mg/kg, group D with gastric infusion of KG 4 g/kg and group E with KG 12 g/kg. The treatment lasted 34 days. Serum levels of prolactin (PRL), testosterone (T), follicle-stimulating hormone (FSH), luteinizing hormone (LH) and estrogen (E2) were detected before and after treatment and the differences between groups were compared. [ Results ] Serum PRL and T levels were increased and LH, FSH and E2 levels decreased in group B (P
